Is it necessary to remember all the commands to be given into .dofile to run the test or are they be inserted through some kind of GUI insertion also like in Modelsim simulator,it is not necessary to remember all the macro to open any screen or to run any task ?

No- It is not necessary to remember all the commands as we have always access of the help of commands and user guide for any tool. But main thing is the documents should be handy. You can insert through GUI also. There is not any issue to insert Scan/whatever using GUI. But we just need an overall idea about commands, so you can search easily in the documents.
